Efect of Seedling Ages and Inoculation Durations with Agrobacterium Tumefaien Transformation Frequency of the Wheat Wounded Apical Meristem

Abstract:Four groups of wheat seedlings, with the different seedling ages of 0 d, 1 d, 2 d and 3 d, were pierced on the apical meristems with a needle. The wounded seedlings are inoculated for 30 min, 3 h and 12 h in the solution of Agrobacterium tumefacien with uidA-nptII genes. The results showed that the seedling ages had a little to do with the percentage of uidA gene transient expression in wheat apical meristems, while the inoculation durations had significant influences. The transient expression percentage of the wounded seedlings inoculated for 30 min was up to 31%, showing the significant potential of promoting the transformation percentage by this planta transformation method.
